Abstract: O_2 consumption method assaying cytotoxic activity of 6MP was compared with other 4 methods; dye exclusion method, colonyformation inhibition method, ^3H-thymidihe uptake inhibition method and growth inhibition method using L1210 mouse leukemic cells and 6MP. The most sensitive method for cytotoxicity assay was colony formation inhibition method on an agar plate but at least 10 days are necessary for determination. Dye exclusion method was the simplest but sensitivity was the lowest among 5 methods and 16 hours are required for the evaluation. O_2 consumption method was more sensitive and reliable than dye exclusion method. Moreover, O_2 consumption method was the simplest among 4 methods except dye exclusion method and the results could be obtained within 8 hour-treatment with 6MP. These results suggest that O_2 consumption method is an useful and convenient for evaluating drug sensitivity of leukemic cells.
