Abstract: The isolated hog thyroid chromatin could be separated into the 0.6M NaCl-soluble and -insoluble fractions. A part of HMG proteins was dissociated from the chromatin at 0.35-0.6M NaCl while another part was preferentially distributed in the 0.6M NaCl-insoluble chromatin fraction. A significant amount of HMG2 was contained in the mononucleosome from the insoluble chromatin while HMG2 as well as HMG1 and H1 histone was not detected in the mononucleosome from the whole chromatin. The insoluble chromatin was more sensitive than the whole chromatin to digestions with micrococcal nuclease and DNase I. HMG1 and HMG2 were selectively dissociated from the chromatin with 10mM spermidine or spermine.
